Анотація: We analyzed the peculiarities of lipoxygenase (LOX) localization and activity in organs of two higher vascular cryptogams at the different phenological phases of development. For the first time it was showed that floating fronds of the annual water fern Salvinia natans (L.) contained 13-LOX with рНopt value of 8.0, when submerged ones – 13-LOX (рНopt 8.0) at the initial phenological phases of development and 9-LOX (рНopt 5.5) at the subsequent phases. In sporocarps 9-LOX (рНоpt 5.5) was identified. At the same time in fronds of the perennial land fern Polystichum aculeatum (L.) Roth. we identified 13-LOX with рНopt value of 7.74, and in the rhizome – 9-LOX (рНopt 7.54). The localization pattern and change of LOXs isoform activity in ferns organs in various phenological phases of development allow us to suggest that enzyme may be involved in the regulation of growth processes that ensure adaptation to environmental conditions

Анотація: The aim of our work was to study biological properties of the polymer based on pseudo-polyamino acids GluLa-DPG-PEG600, its ability to bind albumin, as well as its localization in rat body and influence on physio­logical and functional state of rat kidneys and liver. We have found the ability of GluLa-DPG-PEG600 to bind bovine serum albumin (BSA) using electrophoresis in 5% polyacrylamide gel. Structural and functional state of the liver and kidneys of rats after injections of polymer were investigated by histological analysis of organs and determination the activities of alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, gamma-glutamyltransferase and content of cholesterol and creatinine in blood. Our results showed little toxic effect of GluLa-DPG-PEG600 on rat body. Using fluorescent microscopy we have studied polymer in complex with BSA distribution in rat body: after intravenous injection polymer are localized in kidneys and spleen, and after intramuscular – in liver and brain. It has been shown that polymer passed through the blood-brain barrier and are localized in the immune organ – spleen, indicating GluLa-DPG-PEG600 as a potential drug transporter

Анотація: The derivatives of a succinic acid represent active compounds which realize their biological action via the influence on pro-/antioxidant processes and energetic metabolism. We studied the nitrogen oxide metabolism as well as reactions of the proteins and lipids free-radical peroxide oxidation under a sub-chronic effect of anti-diabetically active compounds – succinic acid derivatives and its metabolites. We found that the studied compounds were able to change the intensity of protein- and lipoperoxidation in a liver and blood serum interconnected with changes in the activity of the key antioxidant enzymes. The derivatives of succinic acid proved to inhibit the nitrogen oxide synthase reflected in a decrease in nitrite- and nitrate-anion level in a liver, blood plasma and urine as well as in changes in the oxidation processes in a liver and blood serum. We concluded in stating that the specified changes of pro-/antioxidant and nitrogen oxide homeostasis may serve as assessment criteria of the succinate-containing compounds’ biological action

Анотація: The participation of enzymatic systems carrying out generation and conversion of reactive oxygen species (ROS), in realization of the stress-protective effect of hydrogen sulfide (H2S) on wheat coleoptile cells was investigated. It has been shown that the treatment of isolated coleoptiles with a 100 μM hydrogen sulfide donor sodium hydrosulfide (NaHS) caused a transient enhancement of the generation of superoxide anion radical (O2•–), an increase of hydrogen peroxide content and superoxide dismutase (SOD) activity in them. The increase in ROS generation was eliminated by the inhibitor of NADPH oxidase imidazole, but not by the peroxidase inhibitor sodium azide. Treatment of coleoptiles with SOD inhibitor sodium diethyldithiocarbamate (DDC) enhanced the generation of O2]•– and neutralized the effect of increasing H2O2 content induced by NaHS. One day after treatment with the H2S donor, the generation of ROS decreased to a control level, while the activity of antioxidant enzymes increased markedly and the resistance of coleoptiles to damaging heating­ was increased. These effects of the hydrogen sulfide donor were eliminated by coleoptiles’ treatment with inhibitors of NADPH oxidase (imidazole) and SOD (DDC). It was concluded that both NADPH oxidase, genera­ting O2•– , and SOD, which turns it into H2O2 performing signaling functions, are involved in the formation of a signal that induces protective systems and causes an increase in heat resistance of plant cells

Анотація: The aim of the work was to study the reverse pH-dependent regulation of the enzymatic activity of the catalytic part of ATP synthase (EC 3.6.3.14) of chloroplast – coupling factor CF1. It was shown that the short-term incubation of isolated CF1 in the media with pH 4.5 or 3.5 leads to inactivation of Ca2+-ATPase, which is rapidly (t1/2 ~ 1 min) restored in the medium containing 0.5-10 mM bicarbonate at pH 7.8. After acid treatment, the rate of Mg2+-ATPase reaction was also stimulated in the presence of 1 mM bicarbonate (рН 7.8; 37 °С). The increase in Ca2+– and Mg2+-АТР activity of CF1 associated with the addition of NaHCO3 solution was completely eliminated after the introduction of 50 mM acetazolamide – a specific inhibitor of carbonic anhydrase. The obtained results suggest the existence of the bound bicarbonate in the CF1 structure, which apparently participates in proton transfer.

Анотація: In the experiments on rats subjected to acute hypoxic histochemical nephropathy, caused by sodium nitrite and 2,4-dinitrophenol, fibrinolytic activities of blood plasma, urine, renal cortex, medulla, and papilla after treatment with flocalin – the activator of KATP channels, were studied­. It was shown that in the conditions of acute kidney hypoxic injury flocalin administration resulted in the increase and essential restoration of fibrinolysis in blood plasma diminished under hypoxia, which was due to the growth of non-enzymatic fibrinolysis, whereas in urine and renal medulla the appreciable increase of enzymatic fibrinolytic activity took place. Moreover, the treatment of hypoxic nephropathy animals by flocalin resulted in the marked restoration of kidney ion regulatory and protein excretory functions that proves the positive influence of KATP channels activation on the one of the biochemical mechanisms of acute kidney injury as well as the protective effect of flocalin in relation to tubular cells of nephron. The obtained results testify to the beneficial effects of KATP channels activation in the conditions of acute hypoxic kidneys injury.

Анотація: Dopamine (DA) is produced and released by immune cells. Recent data pointed to DA as a key mediator between the nervous and immune systems. In the present study we tested the hypothesis that peripheral dopaminergic system plays a negative role in ulcerative colitis pathogenesis via the effect on activity of peripheral blood phagocytes. The study was conducted on male Wistar rats (170-200 g). The peripheral dopaminergic system was destroyed by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) injection (20 mg/kg, s.c., 4 times every 2 h). Colitis was induced by 0.1 ml 6% iodoacetamide enema. Rats were subjected to autopsy on the 18th day. We found that MPTP-treated rats had decreased levels of tyrosine hydroxylase, rate-limiting enzyme of DA synthesis, in colon but not in brain. The number and activity of colonic and peripheral blood granulocytes did not significantly differ in saline- and MPTP-treated rats with colitis. The decreased ROS production by monocytes; increased 1.8-fold the number of CD69 (an early activation marker) positive monocytes and 6-fold intensity of CD69 surface expression were observed in MPTP-treated rats vs. saline-treated rats during colitis. The CD14 (the endotoxin coreceptor of phagocytes) surface expression was 2-fold increased in MPTP-treated rats without colitis, but significantly decreased in both saline- and MPTP-treated rats with colitis. We showed for the first time that the destruction of peripheral dopaminergic neurons leads to the improvement of morphological signs of experimental colitis, which might be through the regulatory effect of dopaminergic system on monocytes phenotype and their respiratory burst activity

Анотація: The aim of this study was to evaluate the effects of N-stearoylethanolamine (NSE) on the number of biochemical parameters that are involved in the development of the stress response (corticosterone, catecholamines, testosterone, TBARS, nitric oxide, serotonin), memory state, exploratory activity and antinociceptive response in rats, using a model of chronic social stress. It has been demonstrated that administration of NSE (14 days , intragastrically, 50 mg/kg) after chronic social stress developed (stress was induced by daily agonistic interaction between animals), normalizes plasma content of corticosterone, testosterone, epinephrine, norepinephrine, dopamine, TBARS, nitrite-anion and increases serotonin level that leads to memory state improvement and increase in exploratory activity in rats. The tail-flick latency did not change, that is it remained at the level of stressed animals

Анотація: Degradation of woody components by termites is associated with symbionts inside their digestive tract. In this study, the major compounds were determined in the extract of the termite guts by GC-MS method. Macrotermes gilvus Hagen (worker caste) termites were collected and their dissected guts underwent methanol extraction. It was found that the gut of the termites has an alkaline environment (pH 8.83 ± 0.31) that supports the digestion of lignocellulose biomass and also helps to solubilize phenolic and recalcitrant compounds resul­ting from the depolymerization of woody components. The GC-MS analysis showed that termite guts contained hydrophobic organosilicon components including dodecamethylcyclohexasiloxane, tetradecamethylcyclohexa­siloxane, hexadecamethylcyclooctasiloxane, and octasiloxane, 1,1,3,3,5,5,7,7,9,9,11,11,13,13,15,15-hexa­decamethyl. The guts also contained a phytosterol, which was identified as β-sitosterol. Further analysis of these water-insoluble compounds is needed to reveal their importance in termite digestion

Анотація: Статтю присвячено аналізу науково-практичної діяльності відділу молекулярної біології Інституту біохімії ім. О. В. Палладіна НАН України в контексті історії його розвитку. Наведено найважливіші результати досліджень молекулярних механізмів регуляції гліколізу та експресії генів у злоякісних пухлинах за гіпоксії, ідентифікації ключових транскрипційних факторів регуляції процесів проліферації та ролі альтернативного сплайсингу в регуляції активності різних форм ензиму PFKFB. В останні роки зусилля співробітників відділу спрямовано на дослідження ролі стресу ендоплазматичного ретикулума і регуляції метаболізму та процесів проліферації на рівні експресії генів у репрограмуванні геному. Результати досліджень дозволять з’ясувати молекулярні основи патогенезу різних захворювань та розробити нові ефективні методи їх діагностики, профілактики та лікування
